L-Glutathione (γ-L-Glutamyl-L-cysteinyl-glycine, GSH) is the most abundant intracellular antioxidant in human cells — present at millimolar concentrations in the cytoplasm, nucleus, and mitochondria. It functions through three primary mechanisms: (1) direct free radical scavenging via the sulfhydryl group on its cysteine residue; (2) serving as the essential substrate for glutathione peroxidase (GPx) enzymes that neutralize hydrogen peroxide and lipid hydroperoxides; and (3) Phase II conjugation via glutathione S-transferases (GSTs), which attach glutathione to xenobiotics, drugs, and toxins to facilitate their excretion.
